"Reporters" Indicate Target Analyte Capture (410)

Problem: Need stable, reliable methods to detect and quantify the capture of target analyte species on test beads.

Project Activity Scope (from Technical Overview Chart): Mechanisms through Sensors and Devices

PNNL Solution:

  • Attach fluorescent tags to tether species that will indicate target analyte capture
  • Employ fluorescent nanoparticles ("quantum dots") captured with target analyte
  • Characterize excitation and emission characteristics for target analyte and possible confounders
  • Validate performance for selected target analytes

Capabilities Employed to Meet Client Needs: Surface chemistry, enzyme chemistry, detector physics, electronics, micro fluidics control, electronics systems, quantitative analysis, fluorescent nanoparticle (quantum dot) synthesis and characterization, quantum dot conjugation and attachment, fluorescence spectroscopy, time-resolved fluorescence, time-correlated single-photon counting (TCSPC).
