Detect Early Effects of Environmental Stress (444)

Problem: Detect very early signs of environmental stress on natural organisms, insects, animals, and/or plants.

Project Activity Scope (from Technical Overview Chart): Mechanisms through Instruments.

Graphic representing the process work flow with links to technologies Determine Sensitive Stress Indicator (445) Conceptual Introduction to DNA Basics (450) DNA Array Basics (451) Alternate Methods to Monitor Stress Effects (446) Dual Array Method for Stress Analysis (447) Two-Color Array Method for Stress Analysis (448) Concept for Two-Color Array Mechanism (449)

PNNL Solution:

  • Collected environmental samples for analysis
  • Identified mRNA* indicators of environmental stress effects in target species
  • Identified mRNA bonding fragments for sensor arrays - prepared arrays
  • Analyzed samples using dual array, differential signal method
  • Analyzed samples using single array two-color method
  • analyzed data and assessed impact of stress on target species
  • Identified source, addressed problem

*mRNA - Messenger Ribonucleic Acid chemical fingerprint for a protein product - originally transcribed from a DNA template.

Capabilities Employed to Meet Client Needs: Nucleic acid extraction, array preparation, hybridization, and data analysis/interpretation.
