Proteomics for Biomarker Identification (253)

Problem: Find high specificity biomarkers for pathogen identification to enable development of sensors and antibodies

Unique High Field Fourier Transform Ion Cyclotron Resonance Mass Spectrometer

Project Activity Scope (from Technical Overview Chart): Mechanisms through Instruments

PNNL Solution:

  • Use state-of-the-art FTICR-MS
  • Use PNNL Global Proteomics capability to identify all the proteins from an organism across different culture conditions
  • Perform comparative analysis of pathogenic versus non-pathogenic organisms for unique biomarkers and use results to create organism peptide identification libraries

Capabilities Employed to Meet Client Needs: Fourier Transform – Ion Cyclotron Resonance (FTICR) – mass spectrometry user facility within PNNL-EMSL; protein extraction protocols; ultra high pressure high performance liquid chromatography separations; Sequest database searching; high speed data extraction and analysis algorithms.
