Problem: Develop a method to rapidly extract exploitable biomarkers from difficult to penetrate bacterial endospores

Project Activity Scope (from Technical Overview Chart): Mechanisms through Sensors and Devices

PNNL Solution:

  • Use supercritical fluid extraction (SFE) methods to rapidly extract usable biomarkers from spore shell without requiring cell lysing
  • Adapt to PNNL’s field portable SFE device
  • Low consumables and rapid extraction times
  • Interface extraction process to selective absorbent for sample preparation and analysis processes
  • Unique biomarkers can be easily identified using MALDI-MS and/or fluorescence arrays

Capabilities Employed to Meet Client Needs: Supercritical fluid expertise including methods and development of field portable equipment; spore preparation expertise; MALDI-MS capability for detection of lipid, peptide and protein biomarkers; GC-MS capability for detection of volatile biomarkers.
